Novel strategy for the treatment of heart failure: gene silencing via RNA gene transfer

A new approach to treating cardiac patients involves transferring RNA to the heart cells in order to influence gene expression. While the RNA function that usually comes to mind involves delivery of genetic information to ribosomes for protein manufacture, RNA also plays a role in regulating protein synthesis by influencing gene activity. Research is ongoing into employing RNA interference (RNAi) to silence specific genes, and microRNA (miRNA) modulations to change gene expression patterns. Once proven, the technologies behind target protein enhancement by gene transfer and target protein suppression by RNAi will enable research into the development of a new class of drugs.

Wolfgang Poller, et al. in the Oxford Journal reported that his team has solved the problem of vector targeting in their research into using cardiac-targeted RNAi strategy to control the regulator of cardiac contractility, phospholamban (PLB). Poller refers to RNA as “truly renaissance molecules" and says that "RNAi may provide an efficient, specific, and cheap solution to inhibit target genes.” He reports that “A major challenge for the future will be to identify the targets of miRNAs that participate in the respective pathomechanisms and to understand their mechanistic contribution in detail" but that "Under appropriate conditions, a therapeutic effect may be achieved not only in genetically determined but also in acquired diseases."
