(Figure 1): In this competitive enzyme immunoassay, cortisol present in a sample competes with a fixed amount of cortisol-HRP conjugate for binding sites on a MAB215P coated microtiter plate. Following a wash step to remove excess conju- gate and unbound sample, TMB substrate is added to measure bound HRP enzyme activity. The measured absorbance at 450nm is inversely proportional to the con- centration of cortisol in the sample.
